P082 Is the new modulator affecting global health outcomes over time?

Objectives: Italy started elexacaftor/tezacaftor/ivacaftor (ETI) in July 2021. This study evaluated the global effects of ETI a longitudinal study, assessing patients prior to and for 6 months after starting ETI. Measures focused on “whole person” included neurocognitive side-effects, physical, mental health-related quality life (HRQoL) outcomes. Methods: was prospective observational that enrolled 92 consecutive pwCF, ages 12–59 years (F/M = 46/47; M age 25.4). An ad-hoc measure, based published literature, developed assess common side-effects 4-point Likert scale (e.g., headache, memory, concentration, insomnia, brain fog). were administered within 2 weeks initiating at 1, 3 months: FEV1, BMI, PHQ-9, GAD-7, GI Symptom Tracker (4 scales: Abdominal Symptoms, Stools, Eating Challenges, Adherence Challenges), Symbol Digit Modality Test (SDMT), CFQ-R. Results: As expected, improvements FEV1 BMI observed across time. Neurocognitive reported most frequently headaches (27–30%) memory (15%-20%), endorsed more often by females than males. Cognitive processing SDMT improved significantly months, but performance baseline 1 standard deviation below mean. Few symptoms over Depression, not anxiety, 3-months post-ETI sustained months. HRQoL majority domains, except Body Image Digestion. Improvements significant from 3-months, with decreases some scales 6-months.
